I emailed in response to a craigslist ad for an executive assistant position and received the same e-mail response that others have posted.
Here is what I sent and the reply:

I saw your ad on craigslist for an executive personal assistant. I read it over several times and know that I fit every aspect that is needed. I strive to be efficient and organized in my own life and know that I'd make a great assistant professionally. I have worked as an ambulance dispatcher in the past and if there is one job you need to be on your toes, making critical decisions quickly and coming up with creative solutions to get things done effectively and efficiently that was it. I know that I can do this job and do it well. I would love to have a chance to meet with you about it, I know you will not be disappointed. Attached you'll find my resume. I believe that honesty and integrity are of foremost importance so feel free to ask any questions you may have about me or my experience. Thank you for your time and consideration.
